I. Background

     NGF 是神經(jīng)營(yíng)養(yǎng)因子（NT）家族的成員，該家族還包括腦源性神經(jīng)營(yíng)養(yǎng)因子 （BDNF）、神經(jīng)營(yíng)養(yǎng)因子 3（NT-3）和神經(jīng)營(yíng)養(yǎng)因子 4/5（NT-4/5）。神經(jīng)營(yíng)養(yǎng)因子是發(fā) 育、維持和存活以及記憶形成和儲(chǔ)存的關(guān)鍵調(diào)節(jié)因子。NGF 在成年基底前腦能神經(jīng)元 （BFCN ）的刺激、存活和表型維持中起著關(guān)鍵作用。它在皮質(zhì)和海馬體中合成，并逆行 輸送到 BFCN 細(xì)胞體，觸發(fā)導(dǎo)致維持正常細(xì)胞功能和形態(tài)的過(guò)程。NGF 可以上調(diào)幾種 能標(biāo)志物，即 ChAT 活性、基因表達(dá)和蛋白質(zhì)水平、乙酰合成和釋放以及囊泡乙酰轉(zhuǎn)運(yùn)蛋白（VAChT）的表達(dá)。

     NGF 的生物學(xué)作用與其結(jié)合的受體類(lèi)型密切相關(guān)。NGF 通過(guò)三種不同類(lèi)型受體的作用 發(fā)揮其細(xì)胞作用。它通過(guò)與原肌球蛋白受體激酶 A（TrkA）受體結(jié)合促進(jìn)細(xì)胞存活，但通 過(guò)與 p75 NTR受體和分選蛋白結(jié)合促進(jìn)細(xì)胞凋亡。TrkA 受體是受體激酶家族的一員， 該家族還包括結(jié)合 BDNF 和 NT4/5 的 TrkB，以及優(yōu)先與 NT3 相互作用的 TrkC。TrkA 受 體是 I 型跨膜蛋白的高親和力激酶受體。TrkA 的配體激活導(dǎo)致受體二聚化和不同殘 基的磷酸化，進(jìn)而促進(jìn)不同信號(hào)通路的激活，包括磷脂酶 C-γ （PLC-γ）、絲裂原活化蛋白 激酶 （MAPK） 和磷脂酰肌醇 3-激酶 （PI3K） 通路。
