I. Background

      PVRIG（CD112R），是脊髓灰質(zhì)炎病毒受體(PVR)家族的共抑制受體，在自然殺傷細(xì) 胞（NK）和 T 細(xì)胞上表達(dá)。它與其配體（CD112 或 PVRL2/nectin-2）結(jié)合，抑制 T 細(xì)胞和 NK 細(xì)胞對癌癥的反應(yīng)。因此，CD112R 作為一種新的免疫檢查點抑制劑在癌癥中 具有很高的潛力。

      CD112（cluster of differentiation 112），也被稱為連接蛋白 2 或 PVR 相關(guān)蛋白 2 （PVRL2），是連接蛋白家族的一員，與腫瘤血管生成、生長和轉(zhuǎn)移相關(guān)。CD112 包括一 個具有三個 igg 樣結(jié)構(gòu)域(兩個 IgC 結(jié)構(gòu)域和一個遠(yuǎn)端 IgV 結(jié)構(gòu)域)的細(xì)胞外區(qū)域，一個細(xì)胞 質(zhì)尾部和一個跨膜區(qū)域。CD112 在抗原呈遞細(xì)胞或腫瘤細(xì)胞上很容易檢測到，其高水平表 達(dá)與大多數(shù)癌癥患者的腫瘤進(jìn)展和不良預(yù)后有關(guān)。

      CD112R 與其配體的結(jié)合可招募 SHP1 和 SHP2，從而在 T 細(xì)胞中觸發(fā) TCR/CD28 信號 抑制。PVRIG 還可以通過與 CD226 競爭性結(jié)合其配體而阻斷其傳遞的刺激信號，來發(fā)揮免 疫抑制的效果。這與其同一家族成員 TIGIT 發(fā)揮免疫抑制的作用機(jī)制類似，并且兩者可能 在免疫抑制上產(chǎn)生協(xié)同作用。
